Carlisle, the seat of Nicholas county, was established in 1816 and probably named for Carlisle, Pennsylvania. It replaced Ellisville as county seat in 1816.

Carlisle was incorporated in 1880.

The Carlisle post office opened in 1817.

The population in 2020 was 2,084.
